Mitochondrial DNA (mtDNA) is subject to the accumulation of mutations. To prevent harmful mtDNA mutations from being passed down to subsequent generations, the female germline, the sole transmitter of mtDNA, employs rigorous mtDNA quality control procedures. Our recent RNAi screen in Drosophila, targeting the molecular basis of this process, identified a programmed germline mitophagy (PGM) vital for mtDNA quality control. Upon germ cell entry into meiosis, PGM was observed to commence, potentially due to the hindrance of the mTOR (mechanistic Target of rapamycin) complex 1 (mTORC1). In a surprising turn of events, the PGM process demands the general macroautophagy/autophagy machinery and the mitophagy adaptor BNIP3, but is independent of the canonical mitophagy genes Pink1 and park (parkin), although these are vital for maintaining the integrity of germline mtDNA. We further determined that the RNA-binding protein Atx2 plays a significant role in regulating PGM. First reported here is the identification and implication of a programmed mitophagy event in germline mtDNA quality control, which underscores the Drosophila ovary's usefulness in studying developmentally regulated mitophagy and autophagy in vivo.
On October 4th, 2019, the University of Bergen, in conjunction with the Industrial and Aquatic Laboratory and Fondazione Guido Bernadini, convened a seminar in Bergen, Norway, on 'Severity and humane endpoints in fish research'. The seminar concluded with a workshop, “Establishing score sheets and defining endpoints in fish experiments,” which was held in Bergen on January 28th, 2020. Participants in the seminar were intended to develop a heightened understanding of fish ethics, severity classifications, and humane endpoints in fish research, using farmed salmonids and lumpfish as case studies. To improve the definition of humane endpoints within fish experiments, the workshop aimed to discuss and develop scoring systems for evaluating related clinical signs. Endpoints for fish health cannot be solely predicated on knowledge of diseases and lesions; rather, they should encompass a detailed understanding of the species, its life stage, anatomical structure, physiological mechanisms, general health and behavioral characteristics. In order to underscore the animal's perspective and needs regarding endpoints, we've changed the name of humane endpoints for fish to piscine endpoints. Key insights from the workshop sessions, including practical advice for developing and using score sheets, are outlined in this paper.

Adhering to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses, the systematic review was pre-registered on PROSPERO under ID#127339. The search across eight databases uncovered research articles that evaluated stigma related to abortion. The task of extracting data was undertaken by four researchers, and the accuracy of the extracted data was validated by two reviewers. Using COSMIN guidelines, a thorough assessment of psychometric properties was undertaken.
A comprehensive analysis of 102 articles yielded 21 that reported original procedures for determining abortion stigma's prevalence. The stigma at both the individual and community levels was measured using specific instruments for people who have had abortions.

Although numerous investigations into interhemispheric functional connectivity (FC) using resting-state (rs-) fMRI have been undertaken, the complex interplay of factors underlying the correlated low-frequency rs-fMRI signal fluctuations across homotopic cortices remains. A clear differentiation between circuit-specific FC and global regulations continues to prove elusive. We have developed a bilateral line-scanning fMRI method that detects laminar-specific resting-state fMRI signals from the homologous forepaw somatosensory cortices in rat brains, providing high spatial and temporal resolution. Two distinct, bilateral spectral fluctuation patterns were identified through spectral coherence analysis. Ultra-slow fluctuations (under 0.04 Hz) occurred across all cortical laminae, in contrast to layer 2/3-specific evoked BOLD activity at 0.05 Hz. The 4-second on, 16-second off block design, coupled with resting-state fluctuation analysis at 0.08-0.1 Hz, informed this analysis. click here The L2/3-specific 0.05 Hz signal, as indicated by evoked BOLD signal measurements at the corpus callosum (CC), is possibly linked to neuronal circuit activity triggered by callosal projections, thereby reducing the frequency of ultra-slow oscillations below 0.04 Hz. Analysis of rs-fMRI power variability clustering revealed that the occurrence of L2/3-specific 008-01Hz signal fluctuations is unaffected by the ultra-slow oscillation across varying trials. Our study investigates a biotechnological response curve strategy analyzing bioactive metabolite production in the marine cyanobacterium Spirulina subsalsa, under varying light energy gradients.
